Description

Principle of the assay: human SCGB1A1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for SCGB1A1 has been precoated onto 96-well plates. Standards(E.coli, E22-N91) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for SCGB1A1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human SCGB1A1 amount of sample captured in plate.
Background: SCGB1A1(secretoglobin family 1A member 1) also known as Uteroglobin is a protein that in humans is encoded by the SCGB1A1 gene. SCGB1A1 is the founding member of the secretoglobin family of small, secreted, disulfide-bridged dimeric proteins found only in mammals. This protein is specifically expressed in Clara cells in the lungs. Numerous studies demonstrated that UG is a multifunctional protein with antiinflammatory/ immunomodulatory properties. It inhibits soluble phospholipase A(2) activity and binds and perhaps sequesters hydrophobic ligands such as progesterone, retinols, polychlorinated biphenyls, phospholipids, and prostaglandins. In addition to its antiinflammatory activities, UG manifests antichemotactic, antiallergic, antitumorigenic, and embryonic growth-stimulatory activities
